I am Ph.D. in Immunology and Molecular Biology. In my thesis, I focus on H. pylori infection, inflammation, and gastric cancer. Firstly, I conducted an epidemiological study to assess H. pylori infection and the severity of the gastric lesions. Secondly, I investigated the genetic variations and the serum levels of potent factors implicated in inflammation, tumorigenesis, and tumor regression (TNF-α, TNFR1, and USF1/USF2), in patients with chronic gastritis, gastric precancerous lesions, and gastric cancer. The findings of my thesis report that genes variations and serum levels can be associated with variations in disease susceptibility and can be used as predictive biomarkers for gastric cancer. I have also taken part in other projects within the lab which enabled me to be a co-author of various publications. We conducted a study to assess the virulence of H. pylori strains that circulate in our area, based on the genotyping of two oncogenes cagA and VacA, and their association with gastric carcinogenesis risk. We remarked that virulent strains were associated with a high risk of gastric cancer and precancerous lesions compared to the non-virulent ones. Furthermore, we led a study to compare the sensitivity and the specificity of PCR technique, histological examination, and ELISA test for the detection of H. pylori infection. We remarked that histological examination should be completed by PCR test to give a reliable diagnosis of H. pylori infection. Moreover, we conducted in silico study to evaluate the interaction of a set of flavonoid compounds with H. pylori-CagA and H. pylori-VacA proteins to identify novel therapeutic molecules. This study showed that some compounds may be a potential alternative treatment to suppress the pathogenicity of H. pylori at a time when the antibiotic resistance of H. pylori increases. Currently, we investigated H. pylori antibiotic resistance to monitor the antibiotic resistance curve of H. pylori in our region and guide clinicians to the right antibiotic to decrease eradication failure.
