My thesis work focused on understanding the mechanism of chronic allograft rejection. Transplant recipients development anti-HLA antibodies and this is associated with transplant atherosclerosis, a manifestation of chronic allograft rejection. The primary goal of my Ph.D. thesis was to characterize targets in endothelial cells (ECs), which may be involved in mediating HLA class I-induced antibody mediated allograft rejection. Currently, I am focused on two projects. In the first, I am studying how mammalian target of rapamycin (mTOR) controls the growth of new blood vessels in malignant diseases such as cancer. The data from this project highlights that the SDF-1/CXCR4 axis activates angiogenesis by utilizing a pathway that requires mTORC2 signal transduction. Furthermore, these data present a potential link between mTORC2, metabolism and angiogenesis, which deserves further exploration. In the second project, I am working on the development of a new drug-screening platform for cancer research. I am developing a novel 3D in vitro system where the tumor microenvironment is encapsulated within a bead, which is ideally suited for high-throughput screening of anti-cancer drugs.
