Research Experience Research Associate, The Rockefeller University Adviser – Luciano Marraffini, Ph.D. Project – Biochemical characterization of CRISPR machinery 2012 - Present Developed in vitro assays to characterize the type IIIA multiprotein complex involved in DNA targeting in CRISPR interference Discovered a novel protein complex involved in CRISPR adaptation Used Cas9 to achieve programmable transcription activation/repression Identified the biochemical functions of key Cas proteins As the sole protein biochemist in my current lab, I established our current pipeline for protein expression, purification, and assay. This pipeline involves recombinant expression, FPLC purification, and enzymological assays involving 32P and fluorescence monitoring Supervised graduate students as well as post docs in these fundamentals Presented results in Infectious disease retreat in Rockefeller University, Gordon conference as well as the CRISPR conference 2014, 2015 Graduate Student, Sloan-Kettering Institute 2008-2012 Adviser – Stewart Shuman, M.D, Ph.D. Thesis: Lessons in ligation, structure function studies of a minimal pluripotent DNA ligase. Conducted structure function analysis of minimized eukaryal ATP-dependent DNA ligase using biochemistry and yeast genetic techniques Used stopped-flow and fast quench instrumentation to probe millisecond kinetic processes !Extensively used MATLAB to perform kinetic data analysis Successfully crystallized and solved crystal structures of different proteins.
