I have been trained as a bench researcher in Neuroscience since 2012. For my Ph.D. studies in India, I evaluated beneficial effects of polysialic acid and its synthetic mimetics on axonal growth and neural repair following spinal cord injury in mice. During this time, I learned to perform thoracic spinal cord laminectomies and compression injuries. Also during this period, I gained extensive experience in tissue culture techniques and became proficient in culturing different primary neural cells (astrocytic, cortical, hippocampal, and microglial cells) and neural cell lines (C6 glioma, IMR-32 Neuroblastoma, U-87 MG astroglioma). I developed expertise cellular and molecular biology techniques such as immunohistochemistry, western blotting and co-immunoprecipitation and became proficient in microscopy and imaging techniques; in particular, confocal microscopy, scanning electron microscopy, and live cell imaging during Ph.D. Further, I became interested in the cell-cell interactions that regulate function of the nervous system. Therefore, for my first postdoctoral training in USA (2017), my work focused in examining the role of astrocytic Ca+2 signaling in the structural and synaptic plasticity associated with behavioral alterations in neurodevelopmental disorders such as fragile-X-syndrome. During that time, I learned cranial window implantation and two-photon imaging of astrocytes. I also learned single-cell RNA-seq and plasmid construction, which furthered my repertoire of molecular biology techniques. Having experience in cell-cell interactions, I returned to spinal cord injury research in 2018 in New York, where I studied the role of transcription factors in enhancing transplanted cell survival following spinal cord injury in rats.
